| Background Information | Transient receptor potential cation channel subfamily V member 6 (UniProt Q9H1D0; also known as Alu-binding protein with zinc finger domain, Calcium transport protein 1, Calcium transporter-like protein, CaT-L, CaT-like, CaT1, ECaC2, Epithelial apical membrane calcium transporter/channel CaT1, Epithelial calcium channel 2, TrpV6) is encoded by the TRPV6 (also known as ABP/ZF, CAT1, CATL, ECAC2, ZFAB) gene (Gene ID 55503) in human. TRPV6 belongs to the transient receptor potential (TRP) family of membrane proteins. The human TRPV6 is an 765-amino acid, six-transmembrane protein with both N- and C-termini exposed intracellularly. TRPV6 contains a pore-forming loop domain located between the fifth and sixth transmembrane region. Among all the TRPV family members, TRPV5 and TRPV6 are the two highly Ca2+-selective channels that play pivotal roles in the Ca2+-reabsorption in the kidney and intestine, respectively. TRPV6 is found in the apical brush-border membrane of the intestinal enterocytes where it regulates calcium entry into the cells. TRPV6 expression is also reported in human placenta, pancreas, and prostate gland. Vitamin D treatment of human colon cancer Caco-2 cells and duodenal explants results in upregulated TRPV6 mRNA levels. |
