| Background Information | 8-Nitroguanosine, a highly reactive redox-active nucleic acid derivative, is formed by the action of peroxynitrite. It is a nitrated base of DNA and RNA formed as a result of chronic inflammation. It has been well established that large amount of nitric oxide molecules and superoxide anion, generated as a result of inflammatory response, can cause nitration of guanosine. Such chemically modified nucleotides increase the incidence of mutation during DNA replication. 8-nitroguanosine is considered as a marker of DNA damage related to mutation and cancer. Nitrated guanine nucleosides and nucleotides in the nucleotide pool are shown to contribute to oxidative stress via production of superoxide mediated by various reductases and may disturb or modulate directly various important proteins and enzymes, such as G-proteins and cGMP-dependent enzymes. Clone NO2-52 is a highly specific monoclonal antibody that recognizes 8-nitroguanine and 8-nitroguanosine. However, it does not cross-react with normal nucleotide bases. (Ref.: Ohshima, H., et al.
