| Background Information | ADP-ribosylation factor-binding protein GGA3 (UniProt: Q9NZ52; also known as Golgi-localized, gamma ear-containing, ARF-binding protein 3) is encoded by the GGA3 (also known as KIAA0154) gene (Gene ID: 23163) in human. GGA3 is a monomeric, ubiquitously expressed protein found in trans-Golgi network (TGN) membrane and endosome membrane. It plays a vital role in protein sorting and trafficking between the trans-Golgi network and endosomes. It mediates the ARF-dependent recruitment of clathrin to the TGN and binds ubiquitinated proteins and membrane cargo molecules with a cytosolic acidic cluster-dileucine (AC-LL) motif. Four isoforms of GGA3 have been described that are produced by alternative splicing. It contains a VHS domain (aa 16-146) that functions as a recognition module for sorting signals composed of an acidic cluster followed by two leucines. Its GAT domain (aa 171-298) is responsible for interaction with ARF-GTP, UBC, and RAB GTPase Binding Effector Protein 1 (RABEP1) and prevents ARF-GTP hydrolysis. GGA3 also has a GAE domain (aa 594-715) that binds accessory proteins regulating GGAs function. GGA3 also contains an auto-inhibitory motif (aa 391-395). GGA3 can be phosphorylated by casein kinase 2 and dephosphorylated by protein phosphatase 2A. GGA3 phosphorylation allows the internal AC-LL motif to bind VHS domain and inhibit the recognition of cargo signals. |
