| Background Information | SH2 domain-containing protein 1B (UniProt: O35324; also known as EWS/FLI1-activated transcript 2, EAT-2) is encoded by the Sh2d1b (also known as Eat2, Eat2a, Sh2d1b1) gene in murine species. EAT-2 is a Src homology 2 (SH2) - domain-containing (aa 5-101) intracellular adaptor protein that is related to signaling lymphocytic activation molecule (SLAM) associated protein (SAP). EAT-2 and SAP are expressed in natural killer (NK) cells, and their combined expression appears to be essential for NK cells to kill abnormal hematopoietic cells. NK cells lacking either SAP or EAT-2 display partial defects in NK cell-mediated cytotoxicity toward abnormal hematopoietic cells and more severe defects are observed when NK cells lacking both adaptors. It plays a role in regulation of effector functions of natural killer (NK) cells by controlling signal transduction through CD244/2B4. It is proposed to act as an inhibitor of CD244-mediated NK cell function including cytotoxicity and interferon-gamma production. It is reported to repress natural cytotoxicity and interferon secretion by a mechanism involving tyrosine phosphorylation of its C-terminus. It binds to the phosphorylated receptors CD84, SLAMF1, LY9 and CD244 and does not bind to non-phosphorylated SLAMF1. Mice lacking EAT-2 are reported to have enhanced NK cell activation responses. EAT-2 is expressed at all stages of maturation in NK cells and in macrophages and dendritic cells. EAT-2 is also expressed in spleen, thymus, lung, kidney, heart, colon and small bowel.
