| Background Information | T-cell differentiation antigen CD6 (UniProt: Q61003; also known as CD6) is encoded by the Cd6 gene (Gene ID: 12511) in murine species. CD6 is a single-pass type I membrane protein that is detected at the immunological synapse the contact zone between antigen-presenting dendritic cells and T-cells. It functions as a costimulatory molecule and promotes T-cell activation and proliferation. After T-cell activation, CD6 is hyperphosphorylated on serine and threonine residues and it is also phosphorylated on tyrosine residues in response to stimulation of the TCR complex. CD6 is predominantly expressed in thymus, lymph node, and spleen. It mediates cell-cell contacts and regulates T-cell responses via its interaction with ALCAM/CD166. It also contributes to signaling cascades triggered by activation of the TCR/CD3 complex. It functions as calcium-dependent pattern receptor that binds and aggregates both Gram-positive and Gram-negative bacteria. It binds both lipopolysaccharide (LPS) from Gram-negative bacteria and lipoteichoic acid from Gram-positive bacteria. It mediates activation of the inflammatory response and the secretion of pro-inflammatory cytokines in response to LPS. CD6 is synthesized with a signal peptide (aa 1-16), which is cleaved off to produce a mature form, which has an extracellular domain (aa 17-398), a transmembrane region (aa 399-419), and a long cytoplasmic tail (aa 420-665). |
