| Background Information | ADAR, also known as Nuclear pore protein 96 (NUP96) named after its molecular weight, or often in the literature as its yeast name, Nup145C, is one of them and NUP96 is made from the autocatalytic cleavage of a larger precursor protein named the Nuclear Pore Complex protein Nup98-Nup96, which also happens to be the source of NUP98 another important nuclear pore protein. NUC96 is found on both the nuclear and cytoplasmic sides of the NPC, whereas NUP98 apparently shuttles between the cytoplasmic side and the nucleoplasmic side of the NPC. Nup96 is part of a NP heptameric complex in the nuclear pore complex. Nup96 complexes with Sec13 in the middle of the heptamer complex. The function of the heptamer is to coat the curvature of the nuclear pore complex between the inner and outer nuclear membranes. Nup96 is predicted to be an alpha helical solenoid. The interaction between Nup96 and Sec13 is at the point of curvature in the heptameric complex. Interestingly, NUP96 and NUP98 also have a crucial role in the function of the immune system and are up regulated by interferons in response to viral infections. A heterozygous mutation in NUP96 has been shown to affect the expression of interferon-regulated genes and to disrupt the normal function of macrophages, and B and T lymphocytes, with no detectable gross pathological abnormalities in other organs. Moreover, the role of Nup96 in the immune response seems to be evolutionarily conserved, as NUP96 in Arabidopsis thaliana has also been shown to be required for the basal defense and resistance responses mediated by plant immune sensors. |
