| Applications | |
|---|---|
| Application | Anti-Cdk1 Antibody, clone A17.1.1 is a Mouse Monoclonal Antibody for detection of Cdk1 also known as p34cdc2, CDC2 & has been validated in IF, IP, EA, WB, IHC(P). |
| Key Applications |
|
| Application Notes | Frozen Sections Immunofluorescence Immunoprecipitation: (Native/denatured, Protein A; 2μg/mg protein lysate) Immunohistology (Formalin, 2-4μg/mL for 30 min at RT) Paraffin Sections (staining requires antigen retrieval). Western Blotting Kinase Assay Optimal working dilutions must be determined by the end user. p34cdc2, a serine/threonine kinase, is activated by cyclins, presumably by dephosphorylation of tyrosine residues. MAB8878 inhibits the activation of p34cdc2 kinase by cyclins and precipitates an active kinase. |

| Specificity | Recognizes a 34 kDa protein cdk1, identified as cdk1/p34cdc2 protein kinase (a catalytic subunit of Maturation Promoting Factor). Its epitope maps near the C-terminus of protein and its core is thought to be LGTPNNEV. It shows no cross-reaction with cdk2/p32 and other cyclin dependent kinases (cdks). |

| Entrez Gene Number |
|
| Entrez Gene Summary | The protein encoded by this gene is a member of the Ser/Thr protein kinase family. This protein is a catalytic subunit of the highly conserved protein kinase complex known as M-phase promoting factor (MPF), which is essential for G1/S and G2/M phase transitions of eukaryotic cell cycle. Mitotic cyclins stably associate with this protein and function as regulatory subunits. The kinase activity of this protein is controlled by cyclin accumulation and destruction through the cell cycle. The phosphorylation and dephosphorylation of this protein also play important regulatory roles in cell cycle control. |

| UniProt Summary | FUNCTION: SwissProt: P06493 # Plays a key role in the control of the eukaryotic cell cycle. It is required in higher cells for entry into S-phase and mitosis. p34 is a component of the kinase complex that phosphorylates the repetitive C-terminus of RNA polymerase II. SIZE: 297 amino acids; 34095 Da SUBUNIT: Forms a stable but non-covalent complex with a regulatory subunit and with a cyclin. Interacts with DLG7. Isoform 2 is unable to complex with cyclin B1 and also fails to bind to the CDK inhibitor p21. Interacts with catalytically active CCNB1 and RALBP1 during mitosis to form an endocytotic complex during interphase. SUBCELLULAR LOCATION: Nucleus (By similarity). SIMILARITY: SwissProt: P06493 ## Belongs to the protein kinase superfamily. CMGC Ser/Thr protein kinase family. CDC2/CDKX subfamily. & Contains 1 protein kinase domain. |
