| Background Information | Lymphocyte cytosolic protein 2 (UniProt Q13094; also known as SH2 domain-containing leukocyte protein of 76 kDa, SLP-76 tyrosine phosphoprotein, SLP76) is encoded by the LCP2 gene (Gene ID 3937) in human. SLP76 is a signaling adaptor molecule expressed in a variety of hematopoietic cells, including lymphocytes, myeloid cells, and platelets. SLP-76 contains three pYESP motifs (Y113 and Y128) that are phosphorylated by ZAP-70 to produce docking sites for SH2-containing proteins, such as exchange factor Vav1, adaptor Nck, resting lymphocyte kinase Rlk, and inducible tyrosine kinase Itk. In addition, SLP-76 binds to the SH3 domain of phospholipase Cγ1 (PLCγ1) and SLP76-deficient T cells show an impaired PLCγ1 activation and calcium mobilization. SLP-76 is essential for mediating TCR signaling, Lcp2-/- mice exhibit impaired thymic development and their T cells fail to develop beyond stage III at the double-negative stage. SLP76 is also a critical determinant of NK-cell development and the the underlying “missing-self” recognition mechanism that prevents NK cells from destroying cells that express self major histocompatibility complex (MHC) class I molecules. SLP76 contains an N-terminal sterile-α motif or SAM (human a.a. 15-81 and moust a.a. 12-78) and a C-terminal SH2 domain (human/mouse a.a. 422-530). |
