| Background Information | Nuclear receptor subfamily 4 group A member 3 (UniProt: Q92570; also known as NR4A3, Mitogen-induced nuclear orphan receptor, Neuron-derived orphan receptor 1, Nuclear hormone receptor NOR-1) is encoded by the NR4A3 (also known as CHN, CSMF, MINOR, NOR1, TEC) gene (Gene ID: 8013) in human. NR4A3 acts as a transcriptional activator that binds to regulatory elements in promoter regions in a cell- and response element (target)-specific manner. It induces gene expression by binding as monomers to the NR4A1 response element (NBRE) 5-AAAAGGTCA-3 site and as homodimers to the Nur response element (NurRE) site in the promoter of their regulated target genes. Its DNA binding region is located within amino acids 289-364. Isoform alpha of NR4A3 is highly expressed in skeletal muscle and isoform beta is predominantly expressed in skeletal muscle. Lower expression levels have been reported in fetal brain and in placenta. NR4A3 plays a role in the regulation of proliferation, survival and differentiation of many different cell types and also in metabolism and inflammation. It is also shown to negatively regulate myeloid progenitor cell proliferation by repressing RUNX1 in a NBRE site-independent manner. NR4A3 activity is induced by inflammatory stimuli in endothelial cells through an NF-kappa-B-dependent transactivation of the NR4A3 promoter. NR4A3 is reported to modulate feeding behavior and is involved in energy balance by inhibiting the glucocorticoid-induced orexigenic neuropeptides AGRP expression. It also regulates glucose transport by regulating translocation of the SLC2A4 transporter to the cell surface. Mutations in NR4A3 gene have been linked to Ewing sarcoma, a highly malignant, metastatic, primitive small round cell tumor of bone and soft tissue. |
