| Background Information | Histone acetyltransferase KAT7 (EC 2.3.1.48; UniProt Q5SVQ0; also known as Histone acetyltransferase binding to ORC1, Lysine acetyltransferase 7, MOZ, MYST-2, SAS2 and TIP60 protein 2, YBF2/SAS3) is encoded by the Kat7 (also known as Hbo1, Myst2) gene (Gene ID 217127) in murine species. KAT7 is a MYST ( MOZ, Ybf2/Sas3, Sas2, and Tip60) family histone acetyltransferase (HAT) originally characterized as a HAT bound to origin recognition complex 1 (ORC1) and named HBO1. KAT7 displays histone H4-specific acetyltransferase activity with reduced activity toward histone H3. KAT7 plays an essential role in adipogenesis, embryonic development, and the survival of erythroblasts in fetal liver. KAT7 interacts with two JADE1 isoforms, JADE1S and JADE1L, that promote KAT7-mediated acetylation of nucleosomal histones. The KAT7-JADE1 complex shows cell cycle-dependent chromatin shuttling during mitosis via a mechanism linked to JADE1 phosphorylation. Nuclear JADE1S and HBO1 localization correlates with that of cell proliferation marker Ki67 and bulk histone H4 acetylation. KAT7 upregulation has been reported in some human cancers and tumor suppressor p53 expression suppresses KAT7 HAT activity. |
