| Background Information | Zinc finger and BTB domain-containing protein 38 (UniProt: Q8NAP3, ZBTB38) is encoded by the ZBTB38 gene (Gene ID: 253461) in human. ZBTB38 is a transcriptional regulator with bimodal DNA-binding specificity. It binds with a higher affinity to methylated CpG dinucleotides in the consensus sequence 5-CGCG-3, but can also bind to E-box elements (5-CACGTG-3). It can also bind specifically to a single methyl-CpG pair and represses transcription in a methyl-CpG-dependent manner. ZBTB38 protein is reported to play an important role in regulating DNA replication and common fragile sites (CFS) stability in a RBBP6- and MCM10-dependent manner and is shown to repress expression of MCM10, which plays an important role in DNA-replication. It is also believed to be involved in the differentiation and/or survival of late post-mitotic neurons. ZBTB38 protein contains ten C2H2-type zinc fingers and its BTB (Broad-Complex, Tramtrack and Bric a brac) domain is localized in the amino acids 33-100. However, the BTB domain is not required for its activation. ZBTB38 is down-regulated under conditions of endoplasmic reticulum (ER) stress, which promotes ER stress-associated apoptosis in human bone marrow neuroblastoma cells. In mice subjected to spinal cord injury (SCI) a repression of ZBTB38 is observed and this is linked to reduced secondary tissue damage after SCI. (Ref.:Cai, Y., et al. (2017).
