| Background Information | Matrix metallopeptidase 12 (MMP-12) is able to degrade elastin, entactin, laminin-1, fibronectin, type IV collagen as well as insulin B-chain and casein. MMP-12 is structurally similar to the classical MMPs (MMP-1, MMP-3); it contains a propeptide with autoinhibitory cysteine switch site, a well conserved zinc site, hinge region and hemopexin domain. MMP-12 lacks a transmembrane domain and furin cleavage site. The zymogen for MMP-12 is about 54 kDa, and is quickly activated to the 45 kDa form, and this breaks down to cascade of active forms, ending with the common 22 kDa form.Stimulated macrophages produce MMP-12; it has also been found in osteosarcoma cells, synovial fibroblasts and lung fibroblasts. MMP-12 levels in quiescent cells and tissues are minimal (with the exception of macrophages), and mitogen stimulation or protein concentration is often needed to visualize the bands. In addition, cell types differ greatly in the quantity of MMP-12 produced. |

| Entrez Gene Number |
|
| Entrez Gene Summary | Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMPs are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. It is thought that the protein encoded by this gene is cleaved at both ends to yield the active enzyme, but this processing has not been fully described. The enzyme degrades soluble and insoluble elastin. It may play a role in aneurysm formation and studies in mice suggest a role in the development of emphysema. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. |

| UniProt Summary | FUNCTION: May be involved in tissue injury and remodeling. Has significant elastolytic activity. Can accept large and small amino acids at the P1 site, but has a preference for leucine. Aromatic or hydrophobic residues are preferred at the P1 site, with small hydrophobic residues (preferably alanine) occupying P3. COFACTOR: Binds 4 calcium ions per subunit. & Binds 2 zinc ions per subunit. SIZE: 470 amino acids; 54002 Da TISSUE SPECIFICITY: Found in alveolar macrophages but not in peripheral blood monocytes. DOMAIN: The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme. SIMILARITY: Belongs to the peptidase M10A family. & Contains 4 hemopexin-like domains. |
| Molecular Weight | The zymogen for MMP-12 is ~ 54 kDa, and is quickly activated to a 45 kDa form. This breaks down to a cascade of active forms, ending with the common 22 kDa form. |
