| Background Information | Polycomb protein group complexes are responsible for the regulation of hundreds of genes in mammals and insects. These proteins are responsible for the assembly and packaging of chromatin and for the induction of methylation on Histone H3. EZH2 (enhancer of zeste homologue 2), also known as Enx1, is a member of the polycomb group of proteins and is involved in cell cycle regulation. EZH2 represses transcription via trimethylation of histone H3 on Lys27 (H3K27) as indicated by the fact that RNAi-mediated knockdown of EZH2 resulted in a loss of H3K27 trimethylation. The expression of this protein in human tumors is aggressive for hormone refractory prostate cancer. In breast cancer EZH2 expression was increased in malignant tumors and promoted anchorage independent and invasive growth in vitro.. EZH2, also known as ENX-1, is ubiquitously expressed during early embryo genesis, and becomes restricted to the central and peripheral nervous systems and sites of fetal hematopoiesis during later development. In the adult, EZH2 is restricted to the spleen, testis and placenta. EZH2 and BMI-1 genes are co expressed in Reed-Sternberg cells of Hodgkin’s disease. Coexpression of BMI-1 and EZH2 is also associated with cycling cells and degree of malignancy in B-cell non-Hogkin’s lymphoma. EZH2 is involved in the progression of prostate cancer, and has been identified as a marker of aggressive breast cancer. Two transcript variants encoding distinct isoforms have been identified for this gene. |
