| Background Information | Androgen receptor (UniProt: P15207; also known as Dihydrotestosterone receptor, Nuclear receptor subfamily 3 group C member 4, AR) is encoded by the Ar (also known as Nr3c4) gene (Gene ID: 24208) in rat. AR is a steroid hormone receptor that mediates the action of testosterone and dihydrotestosterone. AR is shown to be predominantly cytoplasmic in its unligated form, but translocates to the nucleus upon ligand-binding. It can also translocate to the nucleus in unligated form in the presence of RACK1. The AR contains three major functional domains - the N-terminal transcriptional regulation domain, the DNA binding domain (DBD; aa 541-614) and the ligand binding domain (aa 651-882). The DBD contains two C4-type zinc fingers (aa 542-562 and 578-602) that recognize specific DNA consensus sequences and facilitate direct DNA binding of the AR to the promoter and enhancer regions of AR-regulated genes. AR is shown to be phosphorylated in prostate cancer cells in response to several growth factors including EGF. This phosphorylation is induced by c-Src kinase. In AR, tyrosine 517 is one of the major phosphorylation sites and an increase in phosphorylation and Src kinase activity are associated with prostate cancer progression. Phosphorylation of AR by Tyrosine kinase non receptor 2 (TNK2) is reported to enhance the DNA-binding and transcriptional activity. AR can also be phosphorylated at serine 61 by Cdk9, which regulates its promoter selectivity and cell growth. (Ref.: Davey, RA., and Grossmann, M. (2016). Clin. Biochem. Rev. 37(1); 3-15). |

| Application | Anti-Androgen Receptor, PG-21, Cat. No. 06-680, is a rabbit polyclonal antibody that detects Androgen receptor and is tested in ChIP-seq, ChIP, Flow Cytometry, Immunocytochemistry, Immunohistochemistry, Immunoprecipitation, and Western Blotting. |
| Key Applications |
|
| Application Notes | Chromatin Immunoprecipitation (ChIP) Analysis: 1 µg from a representative lot detected Androgen Receptor in Sonicated chromatin prepared from hormone starved (72H), then induced (1 µM CI-4AS-1) overnight LNCaP cells (~1e5 cell equivalents per IP). Immunocytochemistry Analysis: A representative lot from an independent laboratory detected Androgen Receptor in MCF-7 cells stabily transfected with Androgen Receptor (Ghatge, R. P., et al. (2005). Breast Cancer Res. 7(6): R1036-1050). Immunohistochemistry Applications: A representative lot from an independent laboratory detected Androgen Receptor in embryonic rat tissue, neonatal rat testis tissue, and adult rat testis tissue (Nation, T., et al. (2010). J Pediatr Surg. 46(8): 1539-1543.). ChIP-seq: A representative lot from an independent laboratory immunoprecipitated Androgen Receptor from LNCaP cell lysates under certain treated conditions (Zhao, J. C., et al. (2012). Genome Res. 22(2):322-331.). Immunoprecipitation Analysis: 10 µg from a representative lot immunoprecipitated Androgen Receptor in LNCap cell lysate. Flow Cytometry Analysis: 1 µg/mL from a representative lot detected Androgen Receptor in MCF-7 cells. Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user |
