| Background Information | Interleukin-2 (IL-2) is commonly considered a pro-inflammatory cytokine that stimulated T-cell-dependent immune reactions and control regulatory T cells or Tregs. Structurally, IL-2 is thought to have a 4-helix-bundle-type fold and uses a novel type I cytokine receptor (IL-21R) with the common gamma chain cytokine receptor. IL-2 also may intensify Th1-mediated diseases, including autoimmune arthritis and may also be interesting clinical targets for those diseases. IL-2 has also been implicated as a possible therapy for infection and cancer. Anti-tumor therapies that include a combination of recombinant IL-2 and recombinant interferon alpha have been promising in clinical studies. |

The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli.
