| Background Information | SC-35 is also known as Splicing factor SC35 and Splicing factor, arginine/serine-rich 2. This protein is encoded by the SFRS2 gene and is a member of the splicing factor SR family. SC-35 is involved in pre-mRNA splicing and is found in bodies in the nucleus that are highly enriched in poly(A) RNA. These nuclear bodies are referred to as speckles, SC 35 domains, or splicing factor compartments (SFCs). The purpose of these bodies is still widely disputed. Speckles correspond largely to, if not entirely to, ultra structures called inter-chromatin granule clusters (IGCs). SC3-35 is required for pre-mRNA splicing events and is highly involved in spliceosomal interaction and assembly. SC-35 is necessary for assembly of the ATP-dependent splicing complex. It is also necessary for the ATP-dependent communication between pre-mRNA and U1 and U2 snRNPs. |

It is required for formation of the earliest ATP-dependent splicing complex and interacts with spliceosomal components bound to both the 5- and 3-splice sites during spliceosome assembly. It also is required for ATP-dependent interactions of both U1 and U2 snRNPs with pre-mRNA. Interacts with other spliceosomal components, via the RS domains, to form a bridge between the 5- and 3-splice site binding components, U1 snRNP and U2AF. Binds to purine-rich RNA sequences, either 5-AGSAGAGTA-3 (S=C or G) or 5-GTTCGAGTA-3. Can bind to beta-globin mRNA and commit it to the splicing pathway.
