| Background Information | GMP synthase [glutamine-hydrolyzing] (EC 6.3.5.2; UniProt P49915; also known as Glutamine amidotransferase, GMP synthetase) is encoded by the GMPS gene (Gene ID 8833) in human. De novo biosynthesis of Guanosine 5-monophosphate (GMP) is accomplished by reactions catalyzed by several enzymes. Inositol monophosphate (IMP) dehydrogenase 1 and 2 (IMPDH1 and IMPDH2) convert IMP into xanthosine monophosphate (XMP), GMP synthetase (GMPS) then converts XMP into GMP, while guanosine monophosphate reductase (GMPR) catalyzes the reversible conversion of GMP to IMP to fuel back into both the AMP and GMP synthesis pathways. GMPS is a bifunctional two-domain N-type pyrophosphatase whose N-terminal glutaminase domain (a.a. 27-216) generates ammonia from glutamine, while its C-terminal synthetase domain (a.a. 27-435) adenylylates XMP on the xanthine C2 oxygen, thereby priming XMP for attack by a nitrogen nucleophile to form GMP. Human GMPS activity is subjected to an allosteric regulation and displays positive cooperativity for the substrate XMP. GMPS is normally sequestered in the cytoplasm, but translocates to the nucleus in response to genotoxic stress or nucleotide deprivation. GMPS nuclear translocation plays an important role in USP7-mediated stabilization of p53, where p53 is transferred from MDM2 to the GMPS-USP7 deubiquitylation complex. |
