| Background | Delta-aminolevulinic acid dehydratase (ALAD) catalyzes the second step in heme biosynthesis. ALAD is also an endogenous inhibitor of the 26S proteasome (1). Defects in ALAD cause acute hepatic porphyria, an inherited defect in heme biosynthesis in the liver (2). ALAD activity is inhibited by lead, and defects in ALAD result in sensitivity to lead poisoning (3). Reduced expression of ALAD is associated with poor prognosis in breast cancer patients. Overexpression of ALAD in breast cancer cells negatively regulates proliferation, invasion, and transforming growth factor-β (TGF-β)-mediated epithelial-mesenchymal transition (EMT) (4). |
