| Background | Aquaporin 5 (AQP5) is a plasma membrane water channel whose overexpression in human cancers, such as breast, pancreatic, and colon cancers, has been associated with poor prognosis and metastasis. In colorectal cancer cells, AQP5 has been shown to regulate migration and invasion, as well as expression of epithelial-mesenchymal transition (EMT) factors. Knockdown of AQP5 inhibits the Wnt/β-Catenin pathway (1). Localization of AQP5 is altered in gastric cancer, and AQP5 is a marker for pyloric stem cells (2). In hepatocellular carcinoma (HCC), downregulation of AQP5 inhibits metastasis and EMT through inactivation of the NF-κB signaling pathway, and AQP5 is therefore a potential therapeutic target for HCC (3). Phosphorylation of AQP5 at Ser156, possibly by PKA, activates the Ras signaling pathway in NIH/3T3 cells (4). AQP5 alters cell shape and actin dynamics without inducing EMT in normal epithelial MDCK cells, and this effect likely requires phosphorylation of AQP5 at Ser156 (5). |
