| Background | Early B-cell factor 1 (EBF1), also referred to as COE1, is a pioneering transcription factor that is crucial for defining B-cell lineage. In hematopoiesis, EBF1 promotes the expression of genes that specify B-cell lineage commitment, such as FoxO1, PAX5, and IRF-4, while also repressing the expression of transcription factor genes that promote alternate lineages (1,2). EBF1 has also been reported to play a role in olfactory neuronal cell, adipocyte, and early osteoclast differentiation (3-6). Due to the crucial role of EBF1 in B-cell development, mutations in EBF1 can lead to the development of B-cell acute lymphoblastic leukemia (B-ALL) (7,8). EBF1 expression is also suppressed in some solid tumor cancers, including genomic loss in breast cancer and down-regulation in pancreatic ductal adenocarcinomas (9,10). Lower expression levels of EBF1 also correlate with lower survival rates in patients with cholangiocarcinoma and colorectal carcinoma (CRC), and induced overexpression of EBF1 in CRC cell lines induces cell cycle arrest and apoptosis in vivo and in vitro (11,12). |
