| Background | Tubulin polymerization-promoting protein (TPPP, also known as p25) is a microtubule nucleation factor in oligodendrocytes. As a largely unstructured protein, TPPP/p25 induces microtubule polymerization in vitro (1,2). TPPP/p25 is enriched in oligodendrocytes in the brain and may play a unique role in the structure and function of these unique cells (3,4). Oligodendrocytes extend elaborate microtubule arbors that contact neuronal axon segments to generate a myelin sheath that is critical for proper propagation of axonal action potentials. TPPP/p25 may not only play a critical role in establishing the microtubule network for oligodendrocytes, but may also contribute to the generation of Golgi outposts, which are Golgi found outside of the cell body. Golgi outposts may play a specialized role in building the local cytoarchitecture of oligodendrocytes and other brain cells (5,6). Misregulation of TPPP/p25 is linked to several neurodegenerative diseases, including Parkinson’s disease, and may contribute to disease etiology and/or represent a potential therapeutic target (7). |
