| Background | Terminal uridylyl transferase 4 and 7 (TUT4/7), also known as ZCCHC11 and ZCCHC6 respectively, are enzymes that posttranscriptionally uridylate mRNAs and miRNAs at the 3’ end (1,2). The TUT enzymes can monouridylate double-stranded RNA with a single nucleotide overhang, which in turn creates a recognition site for Dicer to process miRNAs (3). Many of the miRNAs recognized by TUT4/7 are involved in cell differentiation such as the HOX gene cluster (4). TUT4 and TUT7 have been shown to be recruited by Lin28 in stem cells to uridylate and block expression of let-7 miRNA, a microRNA with tumor suppressive qualities (3,5,6). TUT4 and TUT7 play a role in the mRNA decay pathway, where they polyuridylate targeted mRNAs to promote turnover (7). TUT4 and TUT7 have also been implicated in T cell activation, innate immune response, and antiviral defenses (8-10). Both TUT proteins have also been shown to prevent retrotransposition of LINE-1 elements, indicating a role in maintaining genomic stability (11). |
