| Background Information | MLK1 (MAP3K9) is a serine–threonine kinase of the Mixed-Lineage Kinase (MLK) family capable of activating the c-Jun N-terminal Kinase (JNK)/Mitogen-Activated Protein Kinase (MAPK) pathway and regulating the other two prinicpal MAPK cascades, p38 and extracellular signal regulated kinase (ERK). Immunoblots and mass spectrometry show MLK is threonine phosphorylated in or near the activation loop. Studies show that MLKs play a role in the TGF-beta1-induced cell death mechanism. |

| UniProt Summary | FUNCTION: Activates the JUN N-terminal pathway. CATALYTIC ACTIVITY: ATP + a protein = ADP + a phosphoprotein. COFACTOR: Magnesium. ENZYME REGULATION: Homodimerization via the leucine zipper domains is required for autophosphorylation and subsequent activation (By similarity). SUBUNIT STRUCTURE: Homodimer (By similarity). TISSUE SPECIFICITY: Expressed in epithelial tumor cell lines of colonic, breast and esophageal origin. PTM: Autophosphorylation on serine and threonine residues within the activation loop plays a role in enzyme activation. Thr-312 is likely to be the main autophosphorylation site. SEQUENCE SIMILARITIES: Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase kinase subfamily. Contains 1 protein kinase domain. Contains 1 SH3 domain. SEQUENCE CAUTION: The sequence AAI11408.1 differs from that shown. Reason: Erroneous initiation. |
