Overview
The most sensitive alkaline phosphatase substrate available for blotting techniques and in situ hybridization chemistry is the 5-bromo-4-chloro-3-indolyl phosphate/p-nitro blue tetrazolium system (BCIP/NBT). Using proprietary production and stabilization procedures, CHEMICON has made available a unique BCIP/NBT reagent that is stable at room temperature and several times more sensitive than existing formulations. For localization of enzyme labeled probes following Western, Northern, Southern or dot blotting, the reagent is added directly to the membrane. Following a 10-15 minute incubation, the reaction is terminated by distilled water rinses. Longer incubations may be employed if necessary, but use of biotin/streptavidin-alkaline phosphatase signaling complexes will negate prolonged reaction times.
